Nasceu em Cedro (Distrito de Paraopeba), hoje Caetanópolis, onde viveu até aos 16 anos.

Era a caçula dos sete filhos do casal Manuel Ferreira de Araújo e Amélia Gonçalves Nunes.

O pai tinha o ofício de serrador (marceneiro) na fábrica de tecidos Cedro & Cachoeira, e era conhecido como Mané Serrador, violeiro e participante das festas de Folia de Reis.

Em 1944, ficou órfã de pai e pouco depois, órfã de mãe, sendo criada por sua irmã Dindinha (Maria Gonçalves) e o irmão José, conhecido como Zé Chilau.

Participou de aulas de catecismo na matriz da Cruzada Eucarística, onde cantava ladainhas em latim no coro da igreja.

Aos 14 anos ingressou como tecelã na fábrica Cedro & Cachoeira. Dois anos depois, mudou-se para Belo Horizonte, indo morar com a irmã Vicentina e o irmão Joaquim. Trabalhou como tecelã, fez o curso normal à noite e, no final de semana, participava dos ensaios do Coral Renascença, na igreja do bairro onde morava.

Cresceu ouvindo Carmem Costa, Ângela Maria e, principalmente, segundo as suas próprias palavras, Elizeth Cardoso e Dalva de Oliveira, das quais sempre teve muita influência, mantendo, no entanto, estilo próprio.

Em 1966 interpretava "Amor quando é amor" (Niquinho e Othon Russo) no filme "Na onda do iê-iê-iê", do diretor Aurélio Teixeira, o que marcaria o início de seu relacionamento com à Jovem Guarda, pois o filme era recheado de números musicais com o pessoal do movimento, destacando-se o cantor Paulo Sérgio, Ed Lincoln, Wanderley Cardoso, Os Vips, Sílvio César, Rosemary, The Fevers, Renato e Seus Blue Caps. No ano seguinte a cantora voltou às telas do cinema, desta vez interpretando a marcha "Carnaval na onda", de José Messias, no filme "Carnaval Barra Limpa", de J.B Tanko. No filme participaram João Roberto Kelly, Emilinha Borba, Ângela Maria, Altemar Dutra, Marlene e Dircinha Batista.

No ano de 1968 fez a sua última atuação em cinema, quando interpretou "Não consigo te esquecer", de Elizabeth, no filme "Jovens Pra Frente", de Alcino Diniz, estrelado por Rosemary, Jair Rodrigues e Oscarito. Após essa gravação parou de flertar com o movimento da Jovem Guarda e passou a cantar samba por influência de Ataulpho Alves e Adelzon Alves.

Ao que se sabe compôs uma única música em parceria com Maurício Tapajós e Paulo César Pinheiro "A flor da pele", que integrou o LP "As forças da natureza".

Pesquisou a música popular brasileira, seus ritmos e seu folclore. Viajou várias vezes para a África, representando o Brasil. Conhecedora das danças e das tradições afro-brasileiras, converteu-se ao candomblé.

Foi uma das cantoras que mais gravou os compositores da Portela, escola para a qual torcia.

Fundou com o marido, o poeta e letrista Paulo César Pinheiro, o Teatro Clara Nunes, no Shopping da Gávea, no Rio de Janeiro.

Faleceu vítima de complicações operatórias decorrentes de uma operação de varizes.

Em 1988, Maria Gonçalves, irmã de Clara Nunes, reuniu em Caetanópolis várias peças do vestuário da cantora, assim como adereços e objetos pessoais e criou uma sala que abriga o acervo de sua obra em um espaço físico com cerca de 120 metros, anexado à creche que leva o seu nome.

Em 2004 a Prefeitura de Caetanópolis inaugurou o Instituto Clara Nunes (anexo à Creche Clara Nunes), ambos coordenados por Maria Gonçalves, irmã velha da que passou a criar a cantora quando esta tinha apenas quatro anos. Empresas como Vallourec & Mannesmann, entre outras, pretende dar apoio à criação de um futuro "Museu Clara Nunes", em memória de sua filha mais ilustre. O museu será no antigo cinema Clube Cedrense, que pertencia à fábrica têxtil Cedro-Cachoeira, no qual a cantora se apresentou pela primeira vez. Ao acervo, coordenado pelo sobrinho da cantora, o músico Márcio Guima, foram incorporadas novas peças.

No ano de 2012, em comemoração ao aniversário de 70 anos, a cantora ganhou um memorial em sua cidade Caetanópolis, em Minas Gerais. Com curadoria de sua irmã mais velha Maria Gonçalves (a Mariquita), o acervo do Memorial Clara Nunes conta com 6.100 peças pessoais e artísticas da artista, cedidas por vários amigos, inclusive, o ex-marido Paulo César Pinheiro. No Rio de Janeiro, neste mesmo ano, foi inaugurado o Mergulhão Clara Nunes, passagem subterrânea que liga os bairros de Madureira e Campinho, na Zona Norte da cidade. Ainda como parte das comemorações dos 70 anos, seu biógrafo, Vagner Fernandes, relançou a segunda edição, revista e aumentada, do livro "Clara Nunes - A Guerreira da Utopia".
